شناسایی مولکولی و تعیین پروفایل مقاومت آنتی بیوتیکی لیستریا مونوسیتوژنز از نمونه های شیر خام، دست کارگران و تجهیزات شیردوشی در شهرکرد

چکیده

لیسرتیا مونوسیتوژنز، عامل بیامری لیسرتیوز، از طریق مرصف موادغذایی آلوده مثل شیر خام، دست یا تجهیزات شیردوشی آلوده به انسان
منتقل میشود. هدف این پژوهش، شناسایی لیسرتیا مونوسیتوژنز در منونههای شیر خام، تجهیزات شیردوشی و دست کارگران گاوداریهای
شهرکرد بود. در این مطالعه ۷۰ منونه شیر خام و ۴۰ منونه از تجهیزات شیردوشی و ۴۰ منونه از دست کارگران جمعآوری و بعد از غنیسازی
اولیه در محیط آبگوشت مغذی بروسال، بر روی محیط پالکام آگار کشت شدند. پس از تایید بیوشیمیایی، DNA کلنیهای مشکوک به روش
جوشاندن استخراج و با پرایمرهای Prs( جنس لیسرتیا( و پرایمرهای 69DG74-DG( گونه لیسرتیا مونوسیتوژنز( به وسیله PCR تکثیر
شدند. مقاومت آنتیبیوتیکی جدایههای تایید شده با روش انتشار از دیسک در برابر پنج آنتیبیوتیک ارزیابی شد. از میان ۱۵۰ منونه، 14
)9/33 درصد( منونه با آزمونهای بیوشیمیای تایید شدند. بر اساس نتایج PCR تعداد ده منونه شیر خام )۶/۶۷ درصد(، یک منونه تجهیزات
شیردوشی )0/67درصد( و یک منونه از دست کارگران )0/67درصد( از نظر جنس لیسرتیا و هشت منونه شیر )۵/۳۳ درصد(، یک منونه تجهیزات
)0/67درصد( و یک منونه دست کارگران )0/67درصد( از نظر گونه لیسرتیا مونوسیتوژنز مثبت بودند. جدایههای تایید شده، بیشرتین مقاومت
را نسبت به آنتیبیوتیکهای سفتی زوکسیم )%90( و اریرتومایسین )%60( و بیشرتین حساسیت را نسبت به آمیکاسین )%90( و آمپیسیلین
)%80( نشان دادند. در نتیجه امکان انتقال لیسرتیا مونوسیتوژنز از تجهیزات شیردوشی و دست کارگران به شیر و برعکس وجود دارد و خطرات
بالقوه لیسرتیوز مرصف کنندگان شیر خام را تهدید میکند.
