تشخیص Neospora caninum در جنین سقط شده گاو با استفاده ازHistopathology ،ELISA ،PCR و Real time PCR

چکیده

زمینه مطالعه: نئوسپوروز گاو توسط Neospora caninum، انگل تک‌یاخته‌ای که باعث عفونت دائمی می‌شود، ایجاد می‌شود. سقط جنین اغلب تنها علامت بیماری است. 
هدف: هدف از این مطالعه بررسی میزان شیوع نئوسپورا کنینوم در برخی گاوداری‌های البرز و قزوین می‌باشد. 
روش کار: در این تحقیق، 105 نمونه از سقط جنین گاو به آزمایشگاه از مهرماه سال 95 تا آبان سال 97 ارسال شد. نمونه‌های دریافتی شامل مغز، قلب و جفت از نقطه نظر آلودگی با N. caninum با استفاده از Histopathology ،ELISA ،PCR و Real Time PCR بررسی گردید. از تکنیک الایزا به‌عنوان آزمون غربالگری اولیه گله مبتلا به سقط و تأیید آن با استفاده از روش‌های آسیب‌شناسی بافتی و مولکولی PCR استفاده گردید.
نتایج: N. caninum باعث نفوذ التهابی تک‌هسته‌ای در اندام‌های مختلف و نکروز کانونی با نفوذ تک‌هسته‌ای در مغز می‌شود، ضایعات غیرچرکی نیز در اندام‌ها نشان می‌دهد عفونت سیستمیک است. میزان عفونت جنین با  N. caninum با هر یک از روش‌های تشخیصی، 6/8 درصد  باElisa ، 9/20 درصد با Real Time PCR  و 7/6 درصد با PCR  بود. 
نتیجه‌گیری نهایی: ارزیابی تکنیک‌ها برای تفسیر این نتایج مهم است. تشخیص سقط جنین ناشی از N. caninum مستلزم شناسایی انگل از طریق تکنیک‌های خاص مانند هیستوپاتولوژی یا روش‌های مولکولی مانند PCR است که از حساسیت بالاتری برخوردار هستند. ترجیحاً با شناسایی ضایعات هیستوپاتولوژیک و انجام آزمون‌های مولکولی به همراه هم می‌توان به نتایج درست‌تری دست پیدا نمود. استفاده از جفت و مهم‌تر از آن، مغز جنین و آزمون‌های PCR و Real Time PCR بهمراه استفاده از هیستوپاتولوژی تشخیص قطعی نئوسپروز را افزایش دهد.

چکیده [English]

Background:Bovine neosporosis is caused by Neosporacaninum, A parasite that causes abortion is often the only observed sign of disease.
Objective: The aim of this study was to investigate the prevalence of Neospora caninum in some Alborz and Qazvin farms.
Methods: in this reaserch 105 samples from bovine abortions were sent to the laboratory, from January 2018 to May 2019. Samples were screened for N. caninum in the brain, heart, kidney, liver, lung, spleen, and placenta, using HistoPatology, ELISA, conventional, PCR and Real Time PCR.. Elisa was used as an initial screen for indication of infectious abortion and the second was histopathology and PCR 
Results: A mononuclear inflammatory infiltrate was predominant in the heart, kidney, liver, lung, brain, and placenta. 
N. caninum causes mononuclear inflammatory infiltrate in various organs and focal necrosis with mononuclear infiltrate in the brain, Non-suppurative lesions were also present in the lung, liver, kidney, and placenta, indicating that N. caninum infection is systemic. The rate of fetal infection with N. caninum, represented separately by each of the diagnostic techniques, was 8/6% with Elisa, 20/9% with Real Time PCR and 6/7% with conventional -PCR.
 Conclusions :The diagnosis of abortion due to N. caninum requires detection of the parasite through techniques that are specific, such as Histopathology, or PCR that are both specific and more sensitive, preferably associated with identification of histopathological lesions placenta and, importantly, fetal brain, should be encouraged to increase the success of the definitive diagnosis of neosporosis by tests of conventionalPCR and Real Time PCR.
