بررسی بیان ژن پروتئین‌ شوک حرارتی 70 (HSP70) مغز، هورمون تیروئیدی T4، صفات ایمنی و عملکردی جوجه‌های گوشتی تغذیه شده با سطوح مختلف خاکشیر (Descurainia sophia) در شرایط تنش گرمایی

چکیده

تنش گرمایی یکی از مهم‌ترین عوامل تأثیرگذار بر عملکرد و سلامت طیور بوده و مواجهه با آن از چالش‌های مهم در صنایع پرورش طیور است. استفاده از ترکیباتی با خواص آنتی‌اُکسیدانی مانند گیاهان دارویی در جهت کاهش آثار تنش گرمایی در طیور توصیه می‌شود. به منظور بررسی اثرات سطوح مختلف خاکشیر بر صفات عملکردی، هورمون تیروکسین و پاسخ HSP70 در جوجه‌های گوشتی در زمان تنش حرارتی حاد، تعداد 300 قطعه جوجه گوشتی یک‌روزه سویه هوبارد در چهار تیمار و پنج تکرار در نظر گرفته شد. گروه‌های آزمایشی شامل: گروه شاهد یا جیره پایه، جیره پایه مکمل شده با نیم درصد خاکشیر، جیره پایه مکمل شده با یک و دو درصد خاکشیر بودند. تنش گرمایی از روز 35 تا 42 دوره پرورش روزانه چهار ساعت با دمای 1±34 درجه سانتی‌گراد اعمال شد. به منظور بررسی پارامترهای هورمونی، سیستم ایمنی و آزمایشات مولکولی در روز 42 پرورش خونگیری انجام و از مغز پنج جوجه از هر تیمار نمونه‌برداری شد. نتایج پژوهش بیانگر این است که سطوح مختلف خاکشیر بر صفات عملکردی و وزن اندام‌های لنفاوی در طیور اثر معنی‌داری نداشت (05/0<P). کمترین میزان هتروفیل و بیشترین میزان لنفوسیت در تیمار خاکشیر 1% و 2% مشاهده شد (05/0>P) و افزایش بیان HSP70 و افزایش غلظت هورمون T4 در تیمار 2% خاکشیر مشاهده شد (05/0>P). به طور کلی اضافه کردن خاکشیر (1% و 2%) به عنوان عامل کاهش دهنده آسیب‌های ناشی از تنش گرمایی محسوب شده و می‌تواند به عنوان افزودنی و مکمل در خوراک طیور مورد استفاده قرار گیرد.

چکیده [English]

Heat stress is one of the most important effective factors on performance and health and exposure to heat stress is one of the major challenges in poultry industry. Therefore, consuming compounds with antioxidant properties such as herbs are recommended to reduce heat stress effects. A total of 300 one-day-old broiler chicks (Hubbard) were randomly divided to four treatments and five replicates due to the investigate of different levels Descurainia sophia on performance, immune system, T4 thyroid hormone and responses of HSP70 in broiler’s brain under acute heat stress condition. Treatments were included: control treatment (basal diet), basal diet with 0.5% Descurainia sophia, basal diet with 1% Descurainia sophia and basal diet with 2% Descurainia sophia. Heat stress were exposed on d 35 to 42 (34±1˚C) for 4 hours. In order to evaluate the hormone parameters, immune system and molecular experiments five chickens from each treatment were slaughtered and blood and brain sampling were performed on d 42 of experiment. The results of experiment showed that the treatments had no significant difference on performance parameters and lymphoid organs weight in poultry (P>0.05). The least ratio of heterophile and the maximum ratio of lymphocyte were observed in 1% and 2% Descurainia sophia treatments and increasing the expression of HSP70 and T4 hormone concentration were observed in 2% Descurainia sophia treatment (P<0.05). In conclusion, the results showed that 1% and 2% Descurainia sophia is considered as a reducing agent in heat stress injuries and could be used as supplement and feed additive in poultry nutrition.
