سکانس کامل سویه IR Clone12 مشتق شده از سویه لاسوتا ویروس نیوکاسل مؤسسه رازی به روش کلونینگ

چکیده

در چند سال اخیر بیماری نیوکاسل در صنعت طیور خسارات گسترده‌ای از جهت تلفات و افت تولید در مزارع پرورش مرغ گوشتی، تخمگذار و مادر گوشتی در کشور ایجاد نموده است. استفاده گسترده از واکسن‌های زنده و غیرفعّال نیوکاسل سطح ایمنی را تا حد زیادی افزایش می‌دهد، امّا هنوز بیماری نیوکاسل مسئله مهمی در کشورهای در حال توسعه نظیر ایران می‌باشد. تحقیق حاضر با هدف تعیین توالی کامل ویروس نیوکاسل سویه Clone12 IR مشتق شده از سویه لاسوتا انجام گرفت. ابتدا ویروس به تخم‌مرغ جنین‌دار SPF 10 روزه تزریق شد.پس از 72 ساعت مایع آلانتوئیک جمع‌آوری شد. مایع آلانتوئیک با کمک سانتریفوژ دور بالاو گرادیانت ساکارز به صورت مناسب تخلیص شد. RNA ویروسی با کیت تجاری مناسب استخراج شد. با استفاده از یک جفت پرایمر اختصاصی (GSP) دو قطعه cDNA ساخته شد. بدون درنظرگرفتن جایگاه ژنی هفت جفت پرایمر طراحی و  ژنوم ویروس با روش PCR آمپلی‌فای شد. هر محصول PCR طولی برابر با (kb 5/2-2) را شامل می‌شد. محصول PCR از روی ژل آگارز یک درصدالکتروفورز استخراج و بر روی پلاسمید pJET1.2 به نسبت سه به یک ligate شد، در ادامه به باکتری TOP10 ترانسفورم شد. پس از تکثیر پلاسمید استخراج گردید. برای اطمینان از کلونینگ موفق آنزیم برش Bgl II استفاده شد. پلاسمیدها برای تعیین توالی به روش سنگر به شرکت تجاری معتبر ارسال شد. در نهایت با استفاده از نرم‌افزار مگا5، تمام 15186 نوکلئوتید تعیین توالی گردید. و در Gene Bank با کد  MH247189 به ثبت رسیده و قابل دسترسی می باشد. ORF شروع و پایان هر ژن مشخص شد.

Newcastle disease (ND) is an important viral disease of poultry causing severe economic losses and drop in egg production in broiler and layers flocks. Despite vaccination with both live attenuated and inactivated NDV strains to immunize birds, ND is still a serious problem in poultry industry especially in developing countries like Iran. plaque purified NDV strain clone IR12 produced by Razi Institute was propagated in 10-day-old SPF emberyonated chicken eggs, then allantoic fluid was harvested. The virus was purified using sucrose-gradient. RNA was extracted and cDNA spanning the whole genome was synthesized using two GSP primers. PCR was run using 7 sets of primers covering the whole cDNA.The PCR products were all gel extracted and ligated to pJET1.2 vector by 3-1 ratios. Successful cloning of each insert was confirmed using restriction enzyme BglII that digests areas flanking the insert in pJET. After confirmation, the inserts were sequenced using Sanger sequencing with pJEt specific primers as well as primers designed to bind to middle regions of each fragment. Sequence of the whole 15186 bp genome was determined by assembling sequences using MEGA 5software. Location of each start (S) and ending (E) gene signals as well as all ORF start and termination codons were determined.
