جداسازی و شناسائی مولکولی قارچ‌های آلوده‌کننده جیره بر پایه سویا و ذرت طیور در منطقه بیرجند

چکیده

به منظور تولید غذای ســالم برای مصرف‌کنندگان، شناخت و کنترل عوامل آلوده‌کننده خوراک دام اجتناب‌ناپذیر است. از دو نوع خوراک طیور، تعداد 212 نمونه جمع‌آوری شد. بعد از آماده‌سازی نمونه‌ها رقت‌های متوالی تهیه گردید و از هر رقت روی محیط کشت غذائی حاوی آگار کشت داده شد. از خوراک دان‌های مرغی منطقه بیرجند خراسان جنوبی آرایه‌های قارچی P. chrysogenum ، P. aurantiogriseum و Penicillium sp. ،Aspergillus flavus و Fusarium proliferatum جداسازی، خالص‌سازی و بر اساس خصوصیات ریخت‌شناسی و مولکولی شناسائی شدند. استخراج DNA ژنومی با استفاده از روش تغییر یافته Chelex 100 و از واکنش زنجیره‌ای پلیمراز برای تکثیر بخشی از ناحیه  rDNA آرایه‌های قارچی استفاده شد. جدایه‌های Rhizopus  بر اساس خصوصیات ریخت شناسی شناسائی شدند. بیشترین و کمترین تعداد پرگنه‌های قارچی مربوط به قارچ پنی‌سیلیوم (16/1 ±17/0) و قارچ فوزاریوم (417/0 ±075/0) بود. بر اساس یافته‌های این پژوهش، کنترل آلودگی خوراک دان به این قارچ‌ها در برنامه‌های مدیریت بهداشت غذائی با روش‌های کنترل محیط انبار و روش‌های بیولوژیک توصیه می‌شود.

چکیده [English]

Fungal growth on poultry feed decreases their nutritional value and constitutes health hazard. To produce healthy food for consumers, recognizing and controlling the contamination of poultry feed is necessary. A total of 212 poultry feed samples including two different feeds were collected and by using dilution plating technique, successive dilutions were prepared and each of dilutions was cultured on solid fungal media cultures. Different fungal species such as Penicillium sp., P.aurantiogriseum, P.chrysogenum, Aspergillus flavus and Fusarium proliferatum were isolated from poultry feed of Birjand area of south Khorasan province and then purified and identified based on morphological and molecular characteristics. DNA extraction were performed by modified Chelex 100 method and Polymerase chain reaction (PCR) method using amplification of  internal transcribed spacer (ITS) region used for molecular identification of fungal taxa. Rhizopus isolates identified based on morphological features. The highest and lowest fungal colonies were Penicillium(0.617±1.16) and Fusarium (0.075±0.417) receptively. Based on this study, controlling the contamination of poultry feed is recommended.
