ارزیابی سوبستراهای سلولی مورد استفاده در تحقیق و تولید فرآورده‌های بیولوژیک از نظر آلودگی به ویروس های BVD 1&2

چکیده

    همزمان با گسترش تولید واکسن بر روی محیط‌های کشت سلولی، شناسایی و از بین بردن ویروس‌های ناخواسته آلوده‌کننده محیط‌های کشت با منشاء حیوانی، و همچنین پیشگیری از آلودگی فرآورده بیولوژیک ناشی ازمحیط‌های کشت سلولی به یکی از دغدغه‌های اصلی تولید‌کنندگان واکسن تبدیل شده است. این مطالعه با هدف بررسی آلودگی احتمالی سوبستراهای سلولی مورد مصرف در تحقیق و تولید فراورده‌های بیولوژیک موسسه تحقیقات واکسن و سرم‌سازی رازی به ویروس‌های BVD1 و BVD2 انجام شد. در این پژوهش از تعداد 44 لاین سلولی با شماره‌های پاساژ و تاریخ های ذخیره‌سازی مختلف در مجموع 100 نمونه مورد بررسی قرار گرفت. پس از استخراج RNA از سلول‌های فوق، ابتدا با استفاده از روش Real Time PCR و بکارگیری یک جفت پرایمر برای شناسایی تمامی Pestivirus، آزمایش غربالگری انجام شد. در این آزمایش از بیوتایپ سایتوپاتیک NADL BVDV1a بعنوان کنترل مثبت استفاده شد. سپس به روش RT- PCR  گونه و زیرگونه آن‌ها تعیین شد. آلودگی در پنج سلول Hek293, IBRS2(BA F/9), RBK, FLK, RK13 دیده شد که همه سلول‌ها بجز IBRS2 در تایپینگ از نوع BVDV1b بودند. تمام سلول‌های مذکور در تحقیق و کنترل کیفی فراورده‌های بیولوژیک کاربرد دارند.

Simultaneously with the expansion of vaccine production on cell culture media, identification and elimination of adventitious viruses infecting culture media of animal origin, as well as prevention of contamination of biological products from cell culture media to one of the main concerns of production Vaccine suppliers. The aim of this study was to investigate the possible contamination of cellular substrates used in research and production of biological products of Razi Vaccine and Serum Research Institute with BVD1 and BVD2 viruses. The address of this investigate was 44 samples from 44 cell lines with different passage numbers and storage dates in a total of 100 samples. After extracting RNA from the above cells, a screening test was performed using Real Time PCR and a pair of primers to identify all Pestivirus. In this experiment, the cytopathic biotype BVDV1a NADL was used as a positive control. Then the species and their subspecies were determined by RT-PCR method. Infection was observed in five cells Hek293, IBRS2, RBK, FLK, RK13, all cells except IBRS2 in BVDV1b typing. All of these cells are used in research and quality control of biological products.
