مقایسه اثر باکتری‌های Lactobacillus plantarum و Lactobacillus rhamnosus جداشده از ماهی شیربت به‌صورت مجزا و ترکیبی بر عملکرد سیستم ایمنی ماهی کپور معمولی

چکیده

در این تحقیق اثر باکتری‌های Lactobacillus plantarum و Lactobacillus rhamnosus بومی جدا شده از روده ماهی شیربت بر پاسخ ایمنی ماهی کپور معمولی (Cyprinuscarpio) ارزیابی‌شد. بدین منظور تعداد 300 قطعه ماهی کپور معمولیبا وزن متوسط 5/11±70 گرم به‌طور تصادفی به پنج گروه تقسیم و به ترتیب با غذای حاوی CFU/ml 107×5 Lactobacillus plantarum ،Lactobacillus rhamnosus CFU/ml 107×5، ترکیب دو باکتری، 250 میلی‌گرم پروبیوتیک تجاری مولتی بهسیل و فاقد پروبیوتیک به مدت 60 روز تغذیه شدند. نمونه خون در روزهای صفر، 30 و 60 از هر گروه اخذ و پارامترهای مختلف ایمنی‌شناسی اندازه‌گیری و مقایسه گردید. همه گروه‌ها در روز 60 آزمایش تحت چالش با باکتری بیماری‌زای Aeromonas hydrophila قرار گرفتند. فعالیت لیزوزیم در گروه‌های تیمار شده با رامنوسوس، ترکیبی و پروبیوتیک تجاری در مقایسه با گروه شاهد به ‌صورت معنی‌داری افزایش‌ یافته بود. میزان میلوپراکسیداز تیمار پروبیوتیک تجاری در روز 30افزایش معنی‌داری داشت. همچنین افزایش معنی‌دار احیای NBT در گروه ترکیبی و پروبیوتیک تجاری دیده شد (05/0>P). قدرت باکتری‌کشی در گروه پلانتاروم و ترکیبی در روز 30 به‌ صورت معنی‌داری بالاتر از گروه شاهد بود. اثر باکتری‌کشی پس از 60 روز در تمامی گروه‌های تیمار شده افزایش معنی‌داری یافت. ایمونوگلوبولین تام در تیمارهای ترکیبی و پلانتاروم به ترتیب در روزهای 30 و 60 دارای افزایش معنی‌دار بود. پس از چالش، گروه جیره ترکیبی، میزان زنده‌مانی بهتری را نسبت به سایر گروه‌ها نشان داد. نتایج به دست آمده نشان داد دو باکتری جدا شده دارای اثرات پروبیوتیکی و تحریک ایمنی مناسب و مقاومت در برابر عوامل بیماری‌زای فرصت‌طلب به ویژه در ترکیب باهم هستند و می‌تواند به عنوان مکمل خوراکی جهت استفاده در جیره خوراکی ماهیان گرمابی مورد استفاد قرار گیرند.

چکیده [English]

In this research, the effect of Lactobacillus plantarum and Lactobacillus rhamnosus bacteria isolated from the gut of the domestic fish (Tor grypus) on the immune response of common carp (Cyprinus carpio) were evaluated. For this purpose, 300 juvenile carps with average weight of 70 ± 11.5 gr were randomly divided into five groups and were fed with feed containing Lactobacillus plantarum, 5×107 CFU/ml Lactobacillus rhamnosus, bacteria combination, 250 mg of commercial product of Multi-Behsil probiotic and non-probiotic for 60 days, respectively. Blood samples were taken at days 0, 30 and 60 from each group and immunological parameters were analyzed and compared. All groups were challenged with Aeromonas hydrophila bacteria on day 60 of the experiment. The lysozyme activity in treated groups with rhamnosus, combination and commercial probiotic significantly increased compared with control group. The activity of myeloproxidase in commercial probiotic group had a significant increase on day 30. There was also a significant rise in NBT reduction in the combination and commercial probiotics group (p <0.05). The bactericidal activity in the plantarum and combination group on day 30 was significantly higher than control group. Bactericidal activity increased significantly after 60 days in all treated groups. Total immunoglobulin in combination and plantarumgroup rose significantly on days 30 and 60, respectively. After the challenge, the combination diet showed a better survival rate than other groups. The results showed that the two isolated bacteria had probiotic effects and appropriate immune stimulation, especially asmixture.
