اثر تغذیه‌ای پودر برگ گیاه مورد (Myrtus communis) بر فراسنجه‌های کمی و کیفی اسپرم و عملکرد آنتی‌اکسیدانی منی و خون در قوچ عربی

چکیده

هدف از این تحقیق بررسی تأثیر پودر برگ گیاه مورد با خاصیت آنتی‌اکسیدانی بر پارامترهای کمی و کیفی اسپرم، ظرفیت آنتی اکسیدانی پلاسمای منی و فعالیت آنزیم سوپراکسید دیسموتاز خون قوچ عربی در فصل غیرتولید‌‌مثلی بود. تعداد ۱۲ رأس قوچ عربی بالغ در قالب 3 تیمار آزمایشی و 4 تکرار استفاده شد. تیمارها شامل سطوح پودر برگ گیاه مورد (۰، ۵/۱۲ و ۲۵ گرم در کیلوگرم ماده خشک جیره) بودند. پس از 42 روز از تغذیه دام‌ها، اسپرم‌‌گیری و خون‌‌‌‌گیری به مدت ۴ هفته متوالی انجام و پارامترهای کمی و کیفی اسپرم و توان آنتی‌اکسیدانی تام پلاسمای منی ارزیابی شدند. همچنین، غلظت تستوسترون سرم خون و فعالیت سوپراکسید دیسموتاز پلاسمای خون مورد سنجش قرار گرفتند. سطح ۲۵ گرم پودر برگ مورد بهترین تأثیر در بهبود شاخص‌های اسپرم و منی شامل جنبایی (88/73%)، زنده‌‌مانی (19/74%)، ناهنجاری (10%)، سلامت غشای پلاسمایی اسپرم (38/43%)، حجم منی (91ml/1)، غلظت اسپرم (ml/109 ×39/1)، ظرفیت آنتی اکسیدانی تام پلاسمای منی (μmol/litr 38/109) و فعالیت سوپراکسید دیسموتاز خون (۱۹U/ml/۵۰۰) نسبت به سطح 5/12 گرم پودر برگ مورد و گروه شاهد داشت (05/0>P). غلظت تستوسترون سرم خون تحت تأثیر تیمارها قرار نگرفت. همبستگی مثبت و معنی‌داری میان توان آنتی‌اکسیدانی تام پلاسمای منی و سوپراکسید دیسموتاز پلاسمای خون با فراسنجه‌های کمی و کیفی اسپرم یافت شد (05/0>p). بطور کلی استفاده از پودر برگ گیاه مورد در جیره موجب بهبود فراسنجه‌های اسپرم و آنتی اکسیدانی پلاسمای منی و خون در قوچ عربی شد. بهترین نتیجه با استفاده از سطح 25 گرم در کیلوگرم جیره از این گیاه بدست آمد.

چکیده [English]

The aim of this study was to investigate the effect of different levels of Myrtle leaf powder on quantitative and qualitative parameters of sperm, antioxidant capacity of semen plasma, and blood superoxide dismutase activity (SOD) in Arabi ram in non-breeding season. 12 adult Arabic rams were used in 3 experimental treatments with 4 replicates per treatment. The treatments consisted of 3 levels of Myrtle leaf powder (0, 12.5 and 25 g/kg of DM). After 42 days feeding of experimental treatments, semen samples were collected and blood samples were obtained once a week for 4 consecutive weeks and the quantitative and qualitative parameters of sperm and total antioxidant capacity of semen were measured. Also, blood samples were subjected to measure testosterone level and SOD activity. 25 g/kg of myrtle powder had the best effect on improvement the sperm and semen characteristics include the motility (73.88%), viability (74.19%), morphological defects (10%), plasma membrane integrity (43.38%), semen volume (1.91 ml) and spermatozoa concentration (1.39 × 109/ml) as well as increased the seminal plasma total antioxidant capacity (TAC) (109.38 μmol/litr) and blood plasma SOD (500.19 U/ml) (p < 0.05). The blood sera testosterone concentration was not affected by treatments. A positive and significant correlation was found between the TAC and SOD with quantitative and qualitative parameters of sperm in Arabi ram (p < 0.05). In conclusion, the use of Myrtle leaf powder in diet improved reproductive performance, seminal plasma TAC and blood SOD activity in Arabi ram. The best result was obtained from 25 g/kg of this plant.
