بررسی حضور کمپیلوباکتر در مرغ‌های عرضه‌شده در فروشگاه‌های سطح شهرستان کرمانشاه

چکیده

کمپیلوباکتر به‌ویژه گونه کمپیلوباکتر ژژونی شایع‌ترین علت گاستروانتریت در انسان است. گونه‌های کمپیلوباکتر غالباً به‌میزان زیادی در مجرای روده‌ای طیور کلونیزه می‌شوند. آلودگی با کمپیلوباکتر ژژونی معمولاً به‌وسیله خوردن مرغ نیم‌پز ایجاد می‌شود. هدف از این مطالعه بررسی فراوانی کمپیلوباکتر در لاشه‌های مرغ عرضه‌شده در فروشگاه‌های سطح شهرستان کرمانشاه با استفاده از تکنیک PCR بود. در مجموع 100 نمونه از انتهای کلون مرغ از مغازه‌های عرضه‌کننده مرغ زنده تهیه شد. پس از آماده‌سازی نمونه‌ها، با استفاده از روش فنل-کلروفرم استخراج DNA از نمونه‌ها صورت گرفت. واکنش PCR با استفاده از سه جفت آغازگر اختصاصی Cadf، flaA و hipO صورت گرفت و نمونه‌های مثبت مشخص شدند. از بین 100 نمونه مورد بررسی فراوانی ژن‌های Cadf ، flaA و hipO به ترتیب 30، 31 و 20 درصد بدست آمد. یعنی 31 نمونه از 100 نمونه مورد بررسی به گونه‌های کمپیلوباکتر آلوده بودند (31 درصد) که 20 تای آن‌ها کمپیلوباکتر ژژونی (20 درصد) بودند و 10 نمونه دیگر، گونه‌های کمپیلوباکتر بجز کمپیلوباکتر ژژونی (10 درصد) بودند. 69 نمونه هم از نظر حضور کمپیلوباکتر منفی (69 درصد) بودند. در یک نمونه هم که فقط ژن flaA تکثیر شد، به‌نظر می رسد که ژن Cadf از دست رفته است. بررسی مطالعات نشان داد که میزان آلودگی به گونه‌های کمپیلوباکتر و نیز میزان شیوع آلودگی به گونه کمپیلوباکتر ژژونی در مناطق مختلف ایران و جهان متفاوت است. دلیل این امر توسط محققین مختلف به محل نمونه‌گیری از لاشه، تکنیک‌های مختلف تشخیصی، فصول نمونه‌گیری و غیره نسبت داده شده است. با توجه به نتایج حاصل، اهمیت گوشت طیور به‌عنوان منبع بالقوه عفونت کمپیلوباکتر مشخص شد.

چکیده [English]

Campylobacter (C.) jejuni is the most common cause of gastroenteritis in humans. Campylobacter spp. often colonize in a large amount in poultry guts. C. jejuni infection is usually caused by eating undercooked chicken. The aim of this study was to evaluate the frequency of Campylobacter in chicken carcasses presented in Kermanshah city stores using the PCR technique. A total of 100 samples from the end of the chicken clone were prepared from live poultry markets. After preparation of the samples, DNAs were extracted by phenol-chloroform method and optical density of the nucleic acid was measured by spectrophotometer. PCR reactions were performed and positive samples were identified. Among 100 samples, frequency of Cadf, flaA and hipO was 30%, 31% and 20%, respectively. It means that 31 out of 100 samples were contaminated with Campylobacter species, 20 of which were C. jejuni (64.5%) and 10 were Campylobacter species other than C. jejuni (35.5%). Sixty nine samples were campylobacter negative, (69%). In one sample that only flaA was amplified, it seems that Cadf gene has been missed. Studies showed that the prevalence of Campylobacter species and Campylobacter jejuni infection is variable in different parts of Iran and the world. The reason has been attributed by different researchers to sampling sites of carcasses, different diagnostic techniques, sampling season and etc. According to the results, the importance of poultry meat was determined as a potential source of Campylobacter infection.
