ارتباط ژن‌های حدت استافیلوکوکوس اورئوس با شمارش سلول‌های بدنی در ورم پستان‌های بالینی و تحت بالینی در استان البرز در فروردین تا شهریور 1393

چکیده

هدف از این مطالعه بررسی حضور برخی ژن‌های مهم در ارتباط با حدت در باکتری‌های استافیلوکوکوس اورئوس جدا شده از موارد ورم پستان بالینی و تحت بالینی و بررسی ارتباط این ژن‌ها با میزان (Somatic Cell Count (SCC یا شمارش سلول‌های بدنی به عنوان یک شاخص التهابی در بافت پستان بود. برای این منظور حضور شش ژن مهم حدت hla ،agrIII ،SpaA ،coa ،clfA و hlb به ترتیب مربوط به عامل توده‌ای‌کننده، کواگولاز، ناحیه X کدکننده پروتئین A، ژن تنظیم کننده فرعی، همولیزین A و همولیزین B، با استفاده از روش PCR بررسی شد. میزان فراوانی این ژن‌ها تعیین شد و با میزان SCC مقایسه شد. بعد از آنالیز ارتباط معنی‌دار بین ژن‌های حدت و افزایش SCC مشاهده شد (05/0 ≥ p). طبق نتایج، شش پروفایل مربوط به ورم پستان‌های بالینی بود و تمامی این شش پروفایل مربوط به ورم پستان بالینی، به غیر از یک پروفایل، به صورت انحصاری در محدوده SCC بیش از 5،000،000 قرار داشتند. این موضوع حاکی از این امر است که افزایش حضور ژن‌های حدت باعث افزایش قدرت بیماری‌زایی باکتری استافیلوکوکوس اورئوس و موجب تغییر ورم پستان ازحالت تحت بالینی به بالینی می‌گردد. همچنین این موضوع حاکی از این امر است که هر چقدر سهم حضور ژن‌های حدت در باکتری بیشتر باشد، حدت باکتری افزایش یافته و با ایجاد تخریب و التهاب بیشتر در بافت پستان باعث افزایش SCC در شیر می‌گردد.

The aim of present study was to detect some important virulence genes of Staphylococcus aureus that were isolated from clinical and subclinical mastitis cases and their correlation with Somatic Cell Count (SCC) as inflammatory indicator of udder was found. Presence of six important genes “clfA, coa, SpaA, agrIII, hla and hlb” encoding clamping factor, coagulase, X region of protein A gene spa, accessory gene regulator, hemolysin A and hemolysin B, respectively, were detected using PCR and the results were compared with SCC. After statistical analysis, a significant correlation between virulence genes and increase in somatic cells was observed (P ≤ 0/05). According to the results, the six virulence gene patterns except one, were presented only in the clinical mastitis isolates and all these isolates were corelated to SCC above 5000000 cell/ml. According to the results, increase in virulence genes that were present in Staphylococcus aureus could lead to change the signes of intra mammary infection from subclinical to clinical mastitis. Furthermore, the results indicates that increasing in the number of virulence genes present in Staphylococcus aureus improves virulence of bacteria to damage mammary tissues, which leads to increase in SCC.
