Introduction: Ostrich, as the largest flightless bird, is susceptible to avian influenza viruses (AIV) including the H5 subtype which posing significant economic risks, particularly in ostrich-farming countries such as South Africa. Objective: In this study, the immunogenicity of a commercial inactivated H5 avian influenza vaccine was evaluated in a flock of adult ostriches using the hemagglutination inhibition (HI) test. Methodology: A serological study was conducted on adult ostriches comprising an equal number of males and females in Khorasan Razavi, Iran. The ostriches were vaccinated twice, at a four-week interval, with the commercial inactivated influenza vaccine (Re-6+Re-8) via subcutaneous injection at a dose of 1 mL per bird. Blood samples were evaluated using the Hemagglutination Inhibition (HI) test two weeks after the second vaccination. Results: The HI titer was higher in the vaccinated group (3.1±0.8) compared to the unvaccinated group (2.3 ± 0.74) (P < 0.05), indicating a stronger immune response. The lowest antibody response observed in both groups was 1, while the highest titer were 3 and 4 in the vaccine and control groups, respectively. Apparently, the vaccine did not elicit a strong immune response, indicating poor immunogenicity and variability in the immune response among the vaccinated ostrich. Conclusion: The weak immune response may be attributed to factors such as vaccine quality, administration issues, or strain mismatch. This study highlights the distinct immune responses in ostriches compared to other poultry species and emphasizes the importance of biosecurity, particularly for farms located near wetlands or bird migration routes. Further studies are needed to improve vaccine efficacy. Avian influenza remains a significant threat to ostrich farming. Enhanced biosecurity, improved vaccination strategies, and ongoing surveillance are essential for managing H5 AIV outbreaks. Collaborative efforts among researchers, veterinarians, and farmers are crucial for the sustainability of the industry.
