Introduction Mycobacterium avium subsp. paratuberculosis (Map) causes Johne’s disease (paratuberculosis) in ruminants. Map represents the slowest growth rate of the cultivable mycobacteria as culture of Map-suspected specimens can take 16 weeks or even more before its colonies are visible. Currently, the IS900 is the most frequently-used genetic marker in PCR-based diagnostic protocols of Map. Specificity of this however, has come into question as IS900-like elements have been reported in genome of other mycobacteria.
Aims Here application of IS900 along with six further genetic loci specific to Map including F57, IS1311, HspX, Map02, ISMav2 and Locus 255 has been described.
Methods PCRs, conducted in singleplex format were of conventional type. Optimization measures on thermal and MgCl2-concentration were employed in a way that a single optimized thermal protocol was applicable for all the markers. This strategy was applied on 30 Iranian bovine (N=24), caprine (N=2) and ovine (N=4) Map isolates representing Alborz, Fars, Isfahan, Zanjan, Qazvin and Tehran provinces.
Results Amplification in all PCR reactions on the whole panels of 30 isolates along with the Map laboratory strain of 316f was successful as the expected PCR products of 560, 704, 608, 211, 278, 507, and 404 bp related to the IS900, F57, IS1311, HspX, Map02, ISMav2 and Locus 255 markers respectively, were produced.
Conclusion We assume in the Iranian environment where IS900 remains the backbone in molecular diagnosis of Map, pairing any of the six extra markers described here with the IS900 element, will improve the capability of the laboratory setting in accurate diagnosis of Map.
