چکیده

محققین جدایه های جدید ویروس آنفلوانزاهای در گردش را ثبت میکنند. برای اثبات جدید بودن عملکرد فنوتیپ ویروس در برابر آنتی سرم استاندارد با آزمایش HI ویژه قابل بررسی میباشد. اگر آنتی سرم استاندارد نتواند ویروس جدید را بطور کامل خنثی کند یعنی تغییرات ژنوتیپی به حدی زیاد بوده که باعث تغییر فنوتیپ شده است. در تحقیقی روش شناسایی فنوتیپی سویه هایی( H9N2 )جدید آنفلوانزا برای اولین بار در ایران طبق دستورالعمل مرکز  CDC  انجام گردید. سه ویروس جدایه آنفلوانزای طیور (سویه (H9N2 بنام های A ، B  و C و ویروس سویه واکسینال (شاخص) بجهت تازه سازی به روش کشت در  تخم مرغ جنین دار SPF   پاساژ داده شد. موسسه رازی سرم پلی کلنال برعلیه ویروس سویه واکسینال آنفلوانزای طیور  H9N2 جدایه اولیه (1377)  را فراهم کرد. بوسیله آنتی ژن های تازه شده و سرم پلی کلنال، آزمایش ویژه سنجش مهار هماگلوتیناسیون انجام گردیده و میزان خنثی شدن ویروس ها در مقابل آنتی سرم پلی کلنال ارزیابی شد. از سه ویروس جدایه تنها نمونه  C در مقایسه با  ویروس شاخص پنج گوده اختلاف نشان داد. نتایج جدایه های A و B تقریبا مشابه ویروس شاخص بودند. برای اطمینان از صحت نتایج آزمایش سه بار تکرار شد و نتایج هر 3 تکرار تقریبا مشابه بود. نتایج این تحقیق صحت کاربردی بودن روش آزمایش ویژه ممانعت از هماگلوتیناسیون را برای شناسایی فنوتیپی جدایه های جدید را اثبات کرد. توصیه می شود آزمایشگاه های رفرانس آنفلوانزای طیور کشور ازاین روش استفاده نمایند.

Researchers record new isolates of circulating influenza viruses. To prove the novelty of the virus phenotype, it is checked against the standard antiserum with a specific HI test. If the standard serum fails to completely neutralize the isolate, the genotypic change has increased to such an extent that it has caused a phenotypic change. In a research, the method of investigation and phenotypic identification of new strains of influenza (H9N2) in Iran was carried out according to the instructions of the CDC center. Three isolates of avian influenza virus (strain (H9N2) named A, B and C and the vaccine strain virus were passaged in SPF embryonated eggs for renewal. (1377) with fresh antigens and polyclonal serum, a special hemagglutination inhibition test was performed and the neutralization rate of viruses against polyclonal antiserum was evaluated .Of the three selected isolates, only sample C showed a difference of five wells compared to the vaccinal virus. The results of isolates A and B were similar to influenza vaccine virus. To ensure the accuracy of the results, the experiment was repeated three times, and the results of all three repetitions were similar. The results of this research proved the applicability of the special hemagglutination (HI) test method for the phenotypic identification of new isolates under the H9N2 type. It is recommended that the country's avian influenza reference laboratories use this method.
