اثر عصاره هیدروالکلی Ceratonia siliqua بر روی کیفیت منی سگ در طول نگهداری در یخچال

چکیده

سرد کردن و ذوب کردن منجر به آسیب‌های اکسیداتیو و تغییرات فیزیکی و شیمیایی در اسپرم می‌شود. هدف از مطالعه حاضر بررسی اثر عصاره هیدروالکلی Ceratonia siliqua (خرنوب) بر روی کیفیت منی سگ در طول نگهداری در دمای 4 درجه سانتی‌گراد است. در این بررسی از 3 قلاده سگ نر سن 7-3 ساله برای جمع‌آوری منی استفاده شد. نحوه اخذ منی بصورت دستی می‌باشد. در این بررسی از قسمت دوم انزال که بخش غنی از اسپرم می‌باشد استفاده شد. نمونه‌های بالای 106 × 200 برای انجام آزمایش استفاده شد. نمونه‌گیری در هفته دو مرتبه انجام پذیرفت و برای از بین بردن تنوع بین نمونه‌ها اسپرم، هر بار همه اسپرم انزالی سگ‌ها با یکدیگر ادغام می‌شوند. اسپرم، توسط سانتریوفوژ با دور 700 g به مدت 5 دقیقه سانتریوفوژ شد و پلاسمای منی دورانداخته شد. پس از رقیق‌سازی، اسپرم ها به 5 گروه : 1- کنترل، 2-کنترل شم، 3-10 µg/mL عصاره خرنوب، 4- 20 µg/mL عصاره خرنوب و 5- 40 µg/mL عصاره خرنوب، تقسیم شدند. عصاره هیدرو الکلی خرنوب در بین گروه‌های درمانی درگروه 20 میکروگرم توانست نسبت به سایر گروه‌های درمانی بیشترین اثر در بهبود پارامترهای اسپرم را داشته باشد و همچنین تفاوت معنی‌داری با گروه‌های کنترل و کنترل شم داشت(۰۵/۰>P). این مطالعه نشان می‌دهد که عصاره هیدروالکلی خرنوب به دلیل داشتن ترکیبات آنتی‌اکسیدانی می‌تواند اسپرم را در برابر آسیب‌های اکسیداتیو ناشی از نگهداری در دمای یخچال محافظت کند.

Cooling and thawing result in oxidative damage, physical and chemical changes in the sperm. The aim of the present study was to evaluate the effect of hydroalcoholic extract of Ceratonia siliqua (carob) on semen quality during storage at 4 ° C. In this study, three male dogs aged 3-7 years were used for semen collection. The method of collection semen is manual. In this study, the second part of the ejaculate, which is rich in sperm, was used. Samples above 200 × 106 were used for the experiments. Sampling was performed twice a week, and to eliminate the diversity of sperm samples, all ejaculate dogs were pooled each time. The pooled sperm was centrifuged by centrifugation at 700 g for 5 minutes and the seminal plasma was discarded. After dilution, spermatozoa were divided into 5 groups: 1) Control, 2) control Sham, 3) 10 µg/ml of carob extract, 4) 20 µg/ml of carob extract, 5) 40 µg/ml of carob extract.
Hydroalcoholic extract of carob had the highest effect on sperm parameters improvement in the 20 µg/ml group compared to the other treatment groups and also had a significant difference with the control and control sham groups (P <0.05). This study demonstrates that Ceratonia siliqua hydroalcoholic extract, due to its antioxidant properties, can protect sperm against oxidative damage caused by refrigerated storage.
