تأثیر بیماری ویروسی سپتی سمی هموراژیک (VHS) بر پارامترهای بیومتری در ماهی قزل آلای رنگین کمان (Oncorhynchus mykiss)

چکیده

پژوهش حاضر به منظور ارزیابی تأثیر بیماری سپتی سمی هموراژیک ویروسی بر صفات بیومتری ماهی قزل آلای رنگین کمان انجام شد. تعداد 281 قطعه ماهی با وزن اولیه 38/3 ± 88 گرم بعد از 18 روز دوره عادت دهی به طور تصادفی به 5 گروه تقسیم شدند. همه گروه‌های آزمایشی با جیره غذایی تجاری تغذیه شدند. گروه اول به عنوان شاهد (بدون تزریق) در نظر گرفته شد، اما گروه‌های دوم تا پنجم به ترتیب با سرم فیزیولوژی سه صدم میلی‌لیتر، سرم فیزیولوژی شش صدم میلی‌لیتر، ویروس سپتی سمی سه صدم میلی‌لیتر و ویروس سپتی سمی شش صدم میلی‌لیتر با غلظت -1 4.5xx104 pfu.ml، تزریق شدند. جهت تشخیص بیماری از تست RT-PCR استفاده شد. مدت دوره‌ی پژوهش 35 روز و رکورد‌برداری صفات وزن بدن و طول ماهیان (طول استاندارد، طول چنگالی و طول کل)، در روز‌های 28 و 35 پرورش انجام شد. مقایسات میانگین این صفات در بین گروه‌های مختلف با استفاده از آزمون توکی (HSD) و نرم‌افزار R انجام شد. در نتایج مربوط به وزن بدن در هفته اول بعد از چالش (روز 28) بین گروه شاهد با گروه‌های تزریق شده با ویروس سپتی سمی در سطح (001/0>P) و همچنین بین گروه تزریق سرم فیزیولوژی و گروه تزریق ویروس سپتی سمی اختلاف معنی‌دار مشاهده شد (05/0>P). در هفته دوم بعد از چالش (روز 35) بین گروه‌های تزریق سپتی سمی با دیگر گروه‌های آزمایشی (گروه شاهد، گروه‌های تزریق سرم فیزیولوژی) برای صفت وزن بدن تفاوت معنی‌دار مشاهده شد (001/0>P). در هفته دوم بعد از چالش بین گروه‌های مختلف آزمایشی برای صفات مرتبط با طول ماهی (طول استاندارد، طول چنگالی و طول کلی) تفاوت معنی‌دار وجود داشت (05/0>P). با توجه به نتایج مشاهده شده بروز بیماری VHS باعث کاهش رشد ماهیان شده و زیان اقتصادی در برخواهد داشت.

To evaluate the influence of VHS disease on rainbow trout biometric traits, this research was conducted. Two hundred eighty-one rainbow trout (88±3.38 g initial weight ) were collected, and after 18 days of adaptation period were randomly divided into five groups. All experimental groups were fed by commercial ration. The first group was selected as a control group (no injection), but the second to fifth groups were injected with 0.03 ml physiological serum, 0.06 ml physiological serum , 0.03 ml VHSV, and 0.06 ml VHSV with the concentration of 4.5x104 pfu.ml - respectively. To diagnose the disease, RT-PCR test was used. The research period was 35 days, and records of body weight, and fish length (standard length, fork length and, total length) were collected on days 28, and 35 of rearing. Mean comparisons of these traits between different groups were conducted using the Tukey test (HSD) and R software. In the results of body weight in the first week after challenge (day 28) significant differences were observed between the control group with VHSV injected groups (p < 0.001) and also between physiological serum with VHSV injected groups (p < 0.05). In the second week after challenge (day 35), there were significant differences (p < 0.001) for body weight between VHSV injected groups with control and physiological injected groups (p < 0.001). In the second week after the challenge for traits related to fish length (standard length, fork length, and total length), there were significant differences between all experimental groups (p < 0.05). Based on the observed results, VHS disease will reduce fish growth, and it will cause economic losses.
