اثر عصارۀ هگزانی استخراج شده از گیاه دارویی کلپوره (Teucrium polium L.i) بر فعالیت تخمیر شکمبه‌ای میکروارگانیسم‌ها در شرایط in vitro

چکیده

کلپوره گیاهی از خانوادۀ نعناعیان بوده که دارای مصارف دارویی فراوانی در طب سنتی بوده ولی اثرات آن بر ویژگی‌های تخمیر شکمبه‌ای تابحال ناشناخته مانده است. از اینرو این آزمایش با هدف بررسی تأثیر عصارۀ هگزانی استخراج شده از گیاه کامل کلپوره با دستگاه سوکسله بر برخی فراسنجه‌های تخمیری میکروارگانیسم‌های شکمبه‌ای (شامل باکتری‌ها، پروتوزوآ و قارچ‌ها) و با کمک یک جیرۀ بالانس شده، در شرایط آزمایشگاهی انجام شد. عصارۀ هگزانی کلپوره با نسبت‌های صفر، 150، 300 و 450 قسمت در میلیون به یک محیط کشت تهیه شده از مایع شکمبۀ گوسفند و بزاق مصنوعی (با نسبت یک به دو) اضافه و سرانجام برخی فراسنجه‌های تخمیری اندازه‌گیری شدند. با افزایش سطح عصاره از صفر به 450 قسمت در میلیون، میزان نیتروژن آمونیاکی و اسیدهای چرب فرار کل (TVFA)، افزایش معنی‌داری (خطی) نسبت به تیمار شاهد نشان دادند ولی سایر موارد شامل pH، فراسنجه‌های تولید گاز (گاز 24 و 48 ساعت، پتانسیل و ثابت نرخ تولید گاز)، گوارش‌پذیری ماده‌ خشک و ماده ‌آلی تغییر ننمودند. ضریب تفکیک‌پذیری (PF)، تودۀ میکروبی تولیدی و بازده تولید تودۀ میکروبی در سطح 450 قسمت در میلیون، از کمترین مقدار برخوردار بودند. نتایج کلی نشان داد که عصارۀ هگزانی کلپوره قادر است برخی فراسنجه‌های تخمیری را در محیط کشت (مانند افزایش نیتروژن آمونیاکی و TVFA)، دستخوش تغییراتی نموده و تا حدودی بهبود بخشد به‌طوری‌که بیشترین این تغییرات در سطح 450 قسمت در میلیون عصاره مشاهده گردید. همچنین به‌نظر می‌رسد که سطوح کمتر از 450 قسمت در میلیون، قادر به تغییر معنی‌دار پارامترهای تخمیری مورد مطالعه در مقایسه با تیمار شاهد نیستند.

Teucrium polium L. is a plant from the Lamiaceae family which is very important in Folk Medicine, however, its effects on fermentation characteristics are still unknown. In this experiment, the effect of hexane oil extracted of Teucrium polium L. by soxhlet device was investigated on some fermentation activities of ruminal microorganism (bacteria, protozoa, and fungi) with a balanced ration, in vitro. The hexane extract of Teucrium polium L. was added to a culture medium prepared from sheep's rumen fluid and artificial saliva (ratio of 1 to 2) with proportions of 0, 150, 300, and 450 ppm, and some fermentation parameters were finally measured. With an increase in the extract level from 0 to 450 ppm, NH3-N and total volatile fatty acids (TVFA) showed a significant increase (linear) compared to the control, but other parameters such as pH, gas production parameters (24 and 48 h gas, potential and constant rate of gas production), degradability of dry matter and organic matter were not differ. The partitioning factor (PF), microbial mass yield, and efficiency of microbial mass yield were lowest at 450 ppm. The overall results showed that the hexane extract of Teucrium polium L. was able to modify and improve some of the fermentation parameters in the culture medium (e.g. increase in ammonia nitrogen and TVFA. Also, the highest changes were observed at 450 ppm. It also seems that levels of less than 450 ppm are not able to significantly change the studied fermentation parameters compared to control.
