ارزیابی واریته تخفیف حدت‌یافته نئوسپورا کنینوم به عنوان واکسن زنده تجربی در موش

چکیده

تک‌یاخته نئوسپورا کانینوم (Neospora caninum)، به عنوان یکی از مهم‌ترین عوامل عفونی بروز سقط جنین در گله‌های گاو در سراسر جهان شناخته می‌شود و از این طریق خسارات بالایی را به صنعت گاوداری در جهان وارد می‌آورد. در حال حاضر بیشترین توجه در تحقیقات جهانی به سوی واکسن زنده معطوف گردیده است؛ در این تحقیق با ارزیابی واریته‌ نئوسپورا کانینوم موسسه رازی در قالب واکسن سعی شده واکسن مؤثرتری برای عفونت نئوسپورا کانینوم یافت شود. 16 موش‌ ماده BALB/c بطور تصادفی به 2 گروه مساوی شامل کنترل و گروه تخفیف حدت‌یافته تقسیم شدند و موش‌ها دو بار با واریته‌ نئوسپورا کانینوم موسسه رازی ایمن شدند. بیست ویک روز بعد از ایمن سازی سرم از تمامی موشها جمع آوری گردید و آزمون‌های الیزا، گاما اینترفرون و PCR نیز بعد از چالش برای بررسی ایمنی ایجاد شده صورت پذیرفت. نتایج بررسی ایمنی هومورال و اینترفرون گاما نشان داد که در گروه ایمن شده پاسخ ایمنی ایجاد شد.کمترین نفوذ ژنوم تک‌یاخته نئوسپورا کانینوم در بافت‌های این گروه را آزمون مولکولی نشان داد. نتایج حاصل از این پژوهش نشان داد که انجام تحقیقات روی سوش و واریته‌های تخفیف حدت یافته و غیر بیماری‌زای نئوسپورا کنینوم در راستای تهیه واکسن زنده محافظت‌کننده امید بخش می‌باشد.

Neospora caninum is one of the most important agent cause abortion in cattle all around the world which can lead in economic loss in cattle herds. Currently, most attention is focused into the live vaccine . So far there is no effective vaccine against neosporosis since the aim of this study was to evaluate the attenuated variety of N. caninum as a potential live vaccine. Sixteen female mice BALB/c were divided randomly into conrtrol and attenuated groups. Twenty-one days after immunization, sera of all groups were taken and subjected for ELISA and gamma interferon (IFN-γ) tests. All mice were challenged by the NC-1 strain of N. caninum. One month after challenge tissue samples were collected for PCR. The results of ELISA and IFN-γ tests indicated that immune response generated in the vaccinated group. The results showed that the attenuated variety of N. caninum stimulated both humeral and cellular immune responses. The results indicated that investigations carried out on non-pathogenic strains and attenuated varieties of Neospora caninum to provide live and protective vaccine is promising.
