The Purpose of this study, were to identify class I integrons and determining antibiotic susceptibility of Pseudomonas aeruginosa isolated from  mastitis. In this cross-sectional study, 150 samples of raw milk were collected from different farms, transported to the laboratory and cultured on specific media. Antimicrobial susceptibility test conducted  by disk diffusion method and MIC (Minimum Inhibitory Concentration) determination test performed by E-test in accordance with CLSI guidelines, in various groups. To identify Class I integron gene PCR test were used. Of total number 150 milk samples, 51 samples (34%) were contaminated with Pseudomonas aeruginosa. Antibiotic test results showed that the highest rate of resistance were observed to Ampicillin, Tetracycline and Kanamycin at the rate of 100%, 94.2% and 94.2%, respectively. Most multiple resistances were observed against Kanamycin, Ampicillin and Tetracycline antibiotics. MIC results showed that isolated bacteria were sensitive to Gentamicin in concentration 0.75-2 μg, Ciprofloxacin at 0.094-0.38 μg and Amikacin at 0.05-4 μg. Results molecular tset showed, one isolate (1.96%) were carried Integrons gene (int-1). Since most integrons genes coding enzymes that inactive antibiotics, presence of Integrons genes in samples of raw milk, treatment infections caused by Pseudomonas aeruginosa wich carring these genes, because of the lack of new antibiotics that have a broad therapeutic range, would creat the major problems. }, keywords = {Pseudomonas aeruginosa,Integron Class I,Antibiotic resistance,Bovine mastitis}, title_fa = {جداسازی باکتری سودوموناس آئروژینوزا و ژن اینتگرون کلاس I از نمونه‌های ورم پستان تحت بالینی گاوهای شیری استان تهران}, abstract_fa = {  باکتری سودوموناس آئروژینوزا از مهم‌ترین پاتوژن‌های فرصت طلب بوده که موجب بروز ورم پستان‌های تحت بالینی مقاوم به درمان در گاوهای شیری می‌باشد. هدف این مطالعه، شناسایی اینتگرون‌های کلاس I در باکتری سودوموناس آئروژینوزای جداشده از ورم پستان و تعیین حساسیت آنتی‌بیوتیکی می‌باشد. در این مطالعه توصیفی-مقطعی، تعداد 150 نمونه شیر خام از دامداری‌های مختلف استان تهران و تعدادی نیز از منطقه شهرکرد جمع‌آوری و بر روی محیط‌های اختصاصی کشت داده شد. آزمایش حساسیت آنتی بیوتیکی با استفاده از روش دیسک دیفیوژن و میزان MIC با E-test مطابق دستورالعمل CLSI با آنتی بیوتیک‌هایی از گروه‌های مختلف انجام گردید. جهت شناسایی ژن‌ اینتگرون کلاس I از آزمون PCR استفاده شد. از مجموع 150 نمونه شیر، 51 نمونه 34 درصد آلوده به باکتری سودوموناس آئروژینوزا بودند. نتایج آزمون آنتی‌بیوگرام نشان داد که بیشترین میزان مقاومت نسبت به آمپی‌سیلین، تتراسایکلین و کانامایسین به میزان 100 درصد، 2/94 و 2/94 درصد به ترتیب مشاهده شد. بیشترین مقاومت چندگانه نسبت به آنتی‌بیوتیکهای کانامایسین/آمپی سیلین/تتراسایکلین مشاهده شد. نتایج MIC‌ مشخص نمود که نمونه‌ها به آنتی‌بیوتیک جنتامایسین بین µg  2-75/0، به آنتی‌بیوتیک سیپروفلوکساسین بین µg  38/0-094/0 و به آنتی‌بیوتیک آمیکاسین بین µg   4-05/0 حساس می‌باشند. نتایج مولکولی نشان داد، 1 جدایه (96/1 درصد) واجد ژن اینتگرون (int-1) می‌باشد. از آنجا که بیشتر اینتگرون‏ها، ژن‏ها و آنزیم‏های تغیر دهنده آنتی‌بیوتیکها را  کد می‏کنند، حضور ژن‏های اینتگرون در نمونه‌های شیر خام درمان عفونت ناشی از باکتری سودوموناس آئروژینوزا حاوی این ژن‏ را به دلیل فقدان آنتی‏بیوتیک جدید که طیف درمان وسیعی داشته باشد، با مشکل جدی روبرو می‌نماید. }, keywords_fa = {سودوموناس آئروژینوزا,اینتگرون کلاس I,مقاومت آنتی‌بیوتیکی,ورم پستان گاو}, url = {https://vj.areeo.ac.ir/article_106059.html}, eprint = {https://vj.areeo.ac.ir/article_106059_e3c4d390ce89d9b67416580bd7be25eb.pdf} }
